Immunocytochemical localization of HCV proteins (NS3 and C)in light and electron microscopies in patients with chronic hepatitis C

Background:Detection of HCV proteins (the non-structural protein,NS3 and the core protein,C)and correlation of the proteins expression with grading and staging in children and adults with chronic type C hepatitis.Material/Methods:The studies were performed on liver biopsies obtained from adults (n=13)and children (n=13)with chronic type C hepatitis.We used routine staining of the preparations for light and electron microscopies,immunocyto- chemical ABC (avidin biotin peroxidase) technique associated with ImmunoMax (biotinylated tyramine) amplification using monoclonal anti-NS3 and anti-C antibodies and,colloidal gold in electron microscopy.Results/Conclusions:Using ImmunoMax amplification, NS3 and C protein were detected mostly in the cytoplasm and also in individual nuclei of hepatocytes at light microscope level. Presence of both proteins in the cytoplasm (mainly in the broadened endoplasmic reticulum channels and in mitochondria)and in hepatocyte nuclei (particularly C protein)was confirmed by electron microscopy.No correlation was noted between expression of the two proteins and grading and staging in both groups of patients although grading and staging were signicantly higher in adults as compared to children (p
